Chapter 2A: Measuring Cancer Anorexia/Cachexia: A Case Study: Introduction

Charles L. Loprinzi, MDThe object of this chapter is to provide the audience with information that might be helpful for the development of trials designed to study potential agents that might be helpful for patients with cancer anorexia and/or cachexia. I have been fortunate to have had the opportunity to work with the North Central Cancer Treatment Group (NCCTG) during a period when a series of such clinical trials have been developed (summarized in the table below):

NCCTG Megestrol Acetate Experience with Cancer
Trial
Patients
Megestrol acetate vs. placebo
133
Megestrol acetate dose/response
342
Megestrol acetate/body composition
12
Megestrol acetate vs. placebo in newly diagnosed small cell lung cancer
243
Megestrol acetate vs. dexamethasone vs. fluoxymesterone
480
Megestrol acetate vs. dronabinol vs. both
445+
Total
1655+

NCCTG Experience with Cancer Anorexia/Cachexia Trials - Other
Trial
Patients
Cyproheptadine vs. placebo
295
Pentoxifylline vs. placebo
70
Hydrazine sulfate vs. placebo/colon cancer
127
Hydrazine sulfate vs. placebo/lung cancer
243
Total
735

In order to best teach this subject, I have provided one of the more recent of these trials along with commentary regarding the reasons that we developed this protocol in the manner in which we did. This protocol completed its patient accrual goals in late 1999.

The entire protocol for this study is available here and includes audio or written comments.

Users may access the protocol two ways. Click here to begin reading through the protocol from the beginning. Alternatively, users may use the links on the left side of the screen to go directly to a specific portion of the protocol and hear commentary on that particular section.
